In presence of DNA lesions\,\nPARP-1 is rapidl y activated and signals the presence of damage by\nattaching ADP-ribose un its to chromatin-associated proteins.\nPoly(ADP-ribosyl)ation may function to organize the recruitment of\nrepair proteins to the DSB by noncovalent pADPr-binding\ninteractions. The central role of poly(ADP-ribose) (pADPr) in the\nrecruitment and assembly of multiprotein repair complexes\nstreng hten the role of DNA-dependent PARPs as regulators of genome\nmaintenance pathways.\nIn spite of the importance of pADPr in the molecular basis of\n DNA damage response\, few pADPr-binding proteins and\npADPr-containing mul tiprotein complexes have been identified so\nfar. Our laboratory is presen tly engaged in proteome-wide\nidentification of proteins with affinity to pADPr. In silico\nprediction of pADPr-binding proteins\, large-scale mass \nspectrometry-based proteome analysis and protein microarray\nanalysis of pADPr-binding proteins were used to establish a\ncomprehensive repertoire of pADPr-associated proteins. The\ninvolvement of putative pADPr-binding proteins in DNA damage\nsignaling is presently under investigation using i n vivo laser\nmicroirradiation-induced DNA damage which permits to evaluat e the\npADPr-dependent recruitment of protein at sites of DNA damage. We\n are currently using Multiple Reaction Monitoring (MRM)\, one of the\nmost selective and sensitive mass spectrometry scan function\, for\nthe quantit ative analysis of pADPr-containing protein complexes\ninvolved in DNA dama ge response and repair.
